Summary

Mosunetuzumab is a humanized full-length T cell engaging bispecific antibody designed to target both cluster of differentiation (CD) 20 (CD20) on B cells and CD3 on T cells. This dual-targeting antibody is designed to redirect T cells to engage and eliminate B cells.1

Proposed Mechanism of Action

Mosunetuzumab is an investigational full length, humanized, IgG1 bispecific antibody that targets both CD3 and CD20.  

Anti-CD20/anti-CD3 bispecifics are proposed to create an immunological synapse between T cells and target B cells.2,3

Concomitant binding to CD3 and CD20

The bispecific concomitantly binds to CD3ε, a component of the TCR complex on the surface of T cells, and to CD20 on the surface of B cells.2,3

Synapse formation

Engagement of both arms of the bispecific (binding to CD3ε and CD20) results in crosslinking of the TCR and formation of an immunologic synapse. T cells are then activated leading to B-cell killing, cytokine release and T-cell proliferation.4
